Measuring equilibrium ultrafast structural and conformational dynamics in bio-molecules: a two dimensional infrared (2D-IR) spectroscopic approach

  Abstract  
  In the recent years, 2D-IR spectroscopy has evolved as one of the ideal experimental techniques to study ultrafast structural and conformational dynamics in proteins and enzymes. This article reviews the 2D-IR experimental methodology and provides a qualitative understanding of the spectral signatures of the various ultrafast dynamic processes in the 2D-IR spectrum. The recent literature relating to the use of the 2D-IR spectroscopic technique on biological macromolecules to estimate the timescales of ultrafast structural motions and to understand the connection of the ultrafast dynamics towards their function has been discussed. The future scope of using this experimental technique in a multi-disciplinary approach would provide molecular level understanding of the structure – dynamics – function relationship of bio-molecules.
