Study on fluorescence property of pancreatic kininogenase and determination of its enzyme activity by fluorimetry

  Abstract  
  A fluorescence spectrometry method to rapidly measure the activity of pancreatic kininogenase (PK) has been established. Under the optimum conditions (pH = 8.0, T = 20?), calibration curve is obtained by measuring fluorescence intensity with a series of different activities of pancreatic kininogenase. A good linear relationship could be obtained between the fluorescence intensity and activity of pancreatic kininogenase in the range of 1.08–21.52 U/mL with a correlation coefficient of 0.9996, and the limit of detection (LOD) was 0.01 U/mL. The relative standard deviations (RSD) were 1.1–3.6% (n = 6) and the recoveries were 90.1%–105.6%. Compared with the pharmacopoeia method, N-benzoyl-L-arginine ethyl-ester (BAEE) method, the proposed method was rapid and convenient operation.
