The application of spectrophotometric stopped-flow method in studies of gas uptake reaction mechanism

  Abstract  
  A few vital issues concerning the application of the spectrophotometric stopped-flow method to study the reaction rate of uptake process for inorganic gases such as CO2 and NO2 are addressed. Two analytical methods of determination of the concentration of carbon and nitrogen dioxides in the chemical models and biological materials are utilized to analyze the mechanism of uptake for the gases studied. Furthermore, it is stressed that determination of carbon dioxide concentrations in biological material (Osteosarcoma 143B cells) using first method described can be used not only to analyze chemical reactions but it can be also a marker of pyruvate's protection efficiency against oxidative stress. Moreover, it is proven that the second method is applicable to assess the concentration of •NO2 in tissue samples from rats with acute pancreatitis induced by L–arginine, where oxidative and nitrosative stress are supposed to play a key role in the pathomechanism of this disease.
